Diagnostic Use

Test to detect PML::RAR alpha transcripts in acute promyelocytic leukaemia (APML) patients with a t(15;17) translocation. This test can be used to determine transcript type at
diagnosis and also for monitoring of minimal residual disease (MRD).

Additional Specimen Information

12 ml peripheral blood in EDTA. 1-3 ml of bone marrow in EDTA or RPMI (anticoagulant).
Transport to the laboratory at ambient temperature within 48 hours of collection to ensure high-quality RNA can be extracted.

Test Method

RNA extraction/storage and nested reverse transcriptase PCR (RT-PCR).
Sanger sequencing is used to determine PML::RAR alpha transcript type in diagnostic samples.
Limit of Detection of the Assay: 1 abnormal cell in 10,000 cells total.
