Interpretation
The standard half-life of itraconazole is 35 hours, with time to steady-state of 7 days.
Reference Intervals
Trough concentrations >1mg/L are effective for treating most fungal infections.
For prophylaxis, trough concentrations >0.5 mg/L are recommended.
Itraconazole toxicity is rare, though the probability of toxicity is increased at plasma concentrations >5 mg/L.
Patients with hypoalbuminaemia have a higher free itraconazole concentration for a given total itraconazole concentration.
Itraconazole inhibits the enzyme CYP3A4, increasing exposure to multiple other drugs metabolised by the same enzmye.
