Advance notice of sample arrival and collection early in the day is required to accommodate the lengthy processing time to stabilise the samples.
Note: Donor and pre-transplant recipient (patient) DNA samples should be tested for informative alleles prior to chimerism analysis. These prior tests incur an additional costs for DNA extraction and informative allele screens by STR analysis. If no pre-transplant sample is available, it is possible to determine pre-transplant informative alleles from DNA isolated from a buccal scraping sample.
24 ml peripheral blood in EDTA. 1-3 ml bone marrow in EDTA or RPMI (with anticoagulant).
Transport to laboratory at ambient temperature within 24 hours of collection.
The AmpFlSTR PCR Amplification Kit is a short tandem repeat (STR) multiplex assay that amplifies 15 tetranucleotide repeat loci (D8S1179, D21S11, D7S820, CSF1PO, D3S1358, THO1, D13S317, D16S539, D2S1338, D19S433, vWA, TPOX, D18S51, D5S818, FGA) and the Amelogenin gender-determining marker in a single PCR amplification.
Short tandem repeats (STR) of 2-4 bp in genomic DNA are often highly variable with regard to the number of repeats present and can be used to distinguish the DNA of one person from that of another. They are useful for monitoring the success of bone marrow transplants by showing whether the cells present in blood or bone marrow are those of the donor or recipient.
