Additional Specimen Information

12 ml peripheral blood in EDTA. 1-3 ml of bone marrow in EDTA or RPMI (anticoagulant), fresh tissue in saline.
Transport to the laboratory at ambient temperature within 48 hours of collection to ensure high-quality RNA can be extracted.

Test Method

This method is adapted from that developed by Chomczynski and Sacchi and uses TRIZOL-LS reagent for the isolation of RNA from biological samples. WBC or tissue are homogenised in TRIZOL-LS, a monophasic solution of phenol and guanidine isothiocyanate. Addition of chloroform and centrifugation separates the organic and aqueous phases, and the RNA is recovered from the aqueous phase by isopropanol precipitation.
